However, the alterations that scientists have made to the newly modified version has reduced its anticoagulant properties by around 90 percent while maintaining its protective benefits.</p><p>Researchers have already tested 3K3A-APC in animal models of multiple sclerosis, amyotrophic lateral sclerosis, and traumatic brain injury with encouraging results.</p><p>The drug is currently under development to treat brain bleeds in people who have experienced a stroke. So far, the drug seems to reduce bleeding in the brain with few side effects or safety concerns.</p><p>As a result of 3K3A-APC's positive performance in other trials, the authors of the current study decided to pit it against Alzheimer's. Lead author Berislav V. Zlokovic, Ph.D. writes:</p><p>"Because of its neuroprotective, vasculoprotective, and anti-inflammatory activities in multiple models of neurological disorders, we investigated whether 3K3A-APC can also protect the brain from the toxic effects of [beta-amyloid] toxin in a mouse model of Alzheimer's disease."</p><p>The team's recent findings feature in the Journal of Experimental Medicine.</p><p>The scientists carried out their study using mice with a number of genetic mutations that research has shown to increase Alzheimer's risk. These animals produce high levels of beta-amyloid and demonstrate both cognitive decline and neuroinflammation.</p><p>As the researchers had theorized, 3K3A-APC reduced the buildup of toxic protein in the brains of these mice.</p><p>Also, the mice did not demonstrate the expected memory deficits that protein buildup produces, and cerebral blood flow was normal. In Alzheimer's disease, signs of inflammation in the brain are common. In mice that received 3K3A-APC, the team noted significantly reduced inflammation.</p><h2>How does 3K3A-APC work?</h2><p>Next, the scientists wanted to understand how this experimental drug was imparting its benefits. They found that 3K3A-APC reduced the amount of an enzyme called beta-secretase 1 (BACE1), which nerve cells create. BACE1 is necessary for the formation of beta-amyloid; without it, plaques cannot form.</p><p>In the past, scientists have tried using various compounds to block BACE1. So far, this has not led to new drugs. For many of us, a case of the flu is little more than unpleasant. However, for individuals in high-risk populations — people who are young, old, or pregnant — flu can lead to serious complications, such as pneumonia.</p><p>Although flu vaccines work well against influenza, they are not effective in preventing all strains, and not everyone has easy access to them.</p><p>Designing a cost-effective and straightforward way to reduce the risk of influenza is a pressing public health concern.</p><p>According to the World Health Organization (WHO), up to 650,000 deaths occur each year globally as a result of influenza.</p><p>The influenza virus primarily targets the epithelial cells of the upper and lower respiratory tract. These cells swarm with communities of bacteria.</p><p>Is it possible that the makeup of these colonies could influence the ability of the influenza virus to set up shop and cause illness?</p><p>Previous studies have shown that manipulating the microbiome can change susceptibility to disease. For instance, one study found that treating mice with oral antibiotics led to increased degeneration of the bronchiole epithelium and a higher risk of death after infection with influenza.</p><p>Also, earlier research by Prof. Foxman showed that people with the influenza virus had increased levels of Streptococcus pneumoniae and Staphylococcus aureus in their nose and throat.</p><p>It seems clear that there is a relationship between the respiratory microbiome and influenza viral infection.</p><p>However, to date, as the authors write, "an association between the nose/throat microbiome and influenza risk has not been demonstrated in human populations." Their latest work, which features in PLOS ONE, sets out to address this issue.</p><h2>Bacterial Protectors</h2><p>To investigate, the scientists used data from the Nicaraguan Household Transmission Study, which took place from 2012–2014.</p><p>The participants were household members of individuals with confirmed influenza. The team followed each of them for 13 days or until they developed flu, whichever came first.</p><p>The current study took data from the 537 individuals who tested negative for influenza at the beginning of the study.</p><p>The researchers took samples of throat and nose bacteria at the beginning of the program. Using DNA sequencing, they were able to build up a picture of the types of bacteria present. They broke this data down into five clusters.</p><p>Having divided the types of bacterial community into five groups, they controlled for other variables, such as smoking, age, cramped living conditions, and flu vaccination.</p><p><strong>"We looked at who had which cluster and whether it makes a difference on whether they got influenza, and it does," Foxman says. "That's the exciting thing about it — it tells us if you have this bacterial community, you have lower risk for getting the flu. That's big news because it really hasn't been shown before."</strong></p><p>These results might help explain why some people are more susceptible to influenza than others.</p><h2>Probiotics for your respiratory system?</h2><p>Proponents of probiotics claim that they encourage so-called "good" bacteria in our guts. It also uncovers novel biological mechanisms that may lead to this neurodegenerative condition. </strong></p><p>About 5.7 million people in the United States are currently living with Alzheimer's disease. A recent report issued by the Centers for Disease Control and Prevention (CDC) predicts that by the year 2060, this number will grow to 13.9 million.</p><p>Genes play an important role in whether a person will develop Alzheimer's or not. Researchers estimate that 60–80 percent of the disease is heritable.</p><p><strong>Therefore, identifying the genes that put some people at risk of Alzheimer's is an important scientific endeavor.</strong></p><p>So far, studies have found more than 20 genetic locations that have links with the condition, but these only explain a small percentage of the genetic variance that accounts for Alzheimer's.</p><p>So, a team of scientists has set out to uncover more genetic risk factors. Professor Danielle Posthuma, from the Vrije Universiteit University in Amsterdam, Netherlands, together with Dr. Ole Andreassen from the University of Oslo, Norway, and Dr. Stephan Ripke from the Broad Institute in Boston, MA, led the new research.</p><h2>Results point to brain's immune cells, lipids</h2><p>Prof. Posthuma and colleagues carried out a genome-wide association study of over 455,000 people of European descent. Some of these individuals had already received a diagnosis of Alzheimer's, while others had a parental history of the disease.</p><p>As a result of this combined analysis, the scientists discovered 29 genome-wide genetic locations that have associations with Alzheimer's, nine of which were new genetic loci.</p><p style="text-align:center"><img alt="Alzheimer's: 9 New Genetic Risk Factors Found" src="http://www.katalay.net/images/lg/e75386e23d26a84375b81191c59433ea.jpg" /></p><p>These newly discovered genes shed some light on possible biological mechanisms that may underlie Alzheimer's. "Specifically," she says, "using single-cell gene expression patterns, we show that genetic changes in genes that are expressed in microglia cells are associated with increased risk for [Alzheimer's disease]."</p><p>"Microglial cells are an important part of the immune system of the brain," the researcher goes on. "This finding suggests that we should widen our focus to also include microglia models when performing functional research in [Alzheimer's], in addition to the conventional approach of neuronal models."</p><p>Iris Jansen, one of the first authors of the paper, adds, "We additionally detect genetic changes in proteins that are involved in lipid components."</p><p>"This link has already been described for the APOE gene," continues Jansen, "but our results show that other lipid proteins might also be genetically affected." The APOE gene is "the strongest genetic risk factor" for Alzheimer's, Jansen explains.</p><p>The findings strengthen "the hypothesis that [Alzheimer's] pathogenesis involves an interplay between inflammation and lipids, as lipid changes might harm immune responses of microglia, thereby affecting the vascular health of the brain."</p><h2>Genes, cognitive reserve, and Alzheimer's</h2><p>Finally, the study sheds light on the link between a person's so-called cognitive reserve and their risk of developing Alzheimer's. Stewart, who is director of the Mount Sinai Diabetes, Obesity, and Metabolism Institute, "because, for the first time, we are able to see rates of human cell beta cell replication that are sufficient to replenish beta cell mass in human beings."</strong></p><p>In earlier work, the team had investigated a small molecule that blocks an enzyme called dual-specificity tyrosine-phosphorylation-regulated kinase 1A (DYRK1A). This molecule led to a beta cell proliferation rate of 1.5 to 3 percent.</p><p>In the new study, the team demonstrated how adding a small molecule from a different class of drug raised the proliferation rate to an average of 5–8 percent. The second drug blocks members of the transforming growth factor beta superfamily (TGFβSF).</p><p>However, while the study has taken an important step by showing that the drug combination can regenerate beta cells fast enough for treatment, there is still some work to do.</p><p>As Dr. Stewart explains, "The next big hurdle is figuring out how to deliver them directly to the pancreas."</p><h2>Diabetes, insulin, and beta cells</h2><p>Diabetes is a disease in which blood glucose rises to harmful levels. Persistent high blood glucose damages blood vessels, nerves, and other body systems. It can lead to vision loss, kidney disease, and heart problems.</p><p>Blood glucose levels rise because of difficulties with producing and using insulin, a hormone that helps the body's cells absorb and use glucose to make energy.</p><p><strong>According to the World Health Organization, estimates suggest that there could be as many as 300 million people worldwide with diabetes by 2025.</strong></p><p>In the United States, diabetes affects about 9.4 percent of the population, which equates to approximately 30.3 million people. The National Institute of Diabetes and Digestive and Kidney Diseases suggest that there are another 84.1 million adults with prediabetes.</p><p>There are two main types of diabetes: type 1 and type 2. Around 90–95 percent of adults with diabetes have type 2.</p><p>In type 1 diabetes, the lack of control of blood glucose occurs because the immune system destroys insulin-producing beta cells in the pancreas.</p><p>Type 2 diabetes usually starts with insulin resistance, a condition in which cells become less effective at using insulin. The pancreas initially compensates by making more insulin, but this is not a long-term solution, and blood glucose levels eventually climb.</p><p>Although the two types have differences, recent studies reveal that type 1 and type 2 diabetes share a major feature: a diminished supply of functioning insulin-producing beta cells.</p><h2>'Achieved previously unattainable rates'</h2><p>Dr. Stewart says that none of the drugs currently available for the treatment of diabetes are sufficiently effective in getting human beta cells to regenerate.</p><p>Researchers are investigating other approaches, such as transplantation of beta cells or the pancreas and treatments that use stem cells to generate new beta cells. However, none of these are in widespread use, Dr. Stewart notes.</p><p>In the earlier work, he and his colleagues had shown that a DYRK1A inhibitor called harmine was able to stimulate a sustained proliferation of adult human beta cells in laboratory cultures.</p><p>In addition, mice with human beta cells in place of their own were able to keep blood sugar levels in the normal range following treatment with harmine.</p><p><strong>This was a major step forward. However, the rate of new beta cell production was too low for the treatment to be effective in humans with diabetes.</strong></p><p>The researchers got the idea of adding a TGFβSF inhibitor to harmine while they were investigating a type of benign tumor that forms in beta cells. This uncovered a novel set of targets for drugs that might enhance beta cell proliferation.</p><p>So, the aim of the recent study was to investigate whether combining the two classes of drug might work — and it did.</p><p>The investigators write that the study reveals how blocking "DYRK1A and TGFβSF signaling induces remarkable and previously unattainable rates of human beta cell proliferation [...] and actually increases human and mouse beta cell numbers."</p><p>The study also explores the mechanisms behind the "remarkable rate of proliferation."
